The 23 rule may result from asymmetric binding of RSS to the RAG-1:RAG-2 dimer. RAG-1 has to rotate to the 12-bp RSS to accommodate its length, the second RAG-1 then prefers binding a 23-bp RSS as not to further distort the conformation of the protein.
